I'm going to buy an new WLAN Router (because it has an built-in VoIP adapter).
I would like to know if it is possible to use my current WRT54G Wlan router as an signal repeater in order to extend the range of my wireless Network.
The new router (connected to WAN) will be placed in the 1. floor and the WRT54G in the 3. floor.

You can set it up using WDS, there are a few guides to this, just do a search.
